Kryxana 200mg Tablet is an oral small-molecule inhibitor classified under cyclin-dependent kinase (CDK) 4/6 inhibitors. It contains the active ingredient ribociclib succinate, equivalent to 200 mg of ribociclib per tablet. This medication is prescribed primarily for adults with hormone receptor (HR)-positive, HER2-negative breast cancer, both in advanced/metastatic and early-stage high-risk settings. It works by halting cancer cell division, helping delay progression or recurrence.

Before starting, consult a physician experienced in oncology. Do not use if you have known allergies to ribociclib, peanuts, soya, or any excipients. Inform your doctor if you have liver, kidney, or heart issues, especially conditions affecting the QT interval. Regular monitoring of blood counts, liver function, and ECG is essential. Pregnancy and breastfeeding are contraindications; effective contraception is required during treatment and for at least 3 weeks after the last dose.

• Advanced or metastatic breast cancer
• Advanced or metastatic endometrial cancer

Ribociclib works by blocking specific proteins (CDK4 and CDK6) that help cancer cells grow and divide. By inhibiting these proteins, the drug prevents tumor cells from multiplying, slowing down disease progression in both early and advanced breast cancer. Taken alongside hormonal therapies, it boosts their effectiveness and reduces the risk of cancer returning after surgery. This dual action improves patient outcomes with manageable side effects.

Always discuss all medications with your healthcare provider before starting this treatment. Serious interactions must be considered:

• Strong CYP3A4 inhibitors (e.g., ketoconazole, clarithromycin, ritonavir): can dramatically increase ribociclib levels, raising the risk of toxicity and QT prolongation. Dose reduction or avoiding combination is advised. 

• Strong CYP3A4 inducers (e.g., rifampicin, St John’s wort): may lower ribociclib exposure, risking reduced effectiveness—avoid these combinations. 

• QT‑prolonging drugs (e.g., antiarrhythmics, haloperidol, moxifloxacin, amiodarone): co-use increases risk of dangerous heart rhythm changes. Baseline and ongoing ECG monitoring is recommended.

• Sensitive CYP3A4 substrates (e.g., midazolam): ribociclib can raise their levels, increasing side effects—dose adjustments may be needed. 

Contraindications include known hypersensitivity to the drug, peanut or soy allergies, pregnancy, and breastfeeding. It is also contraindicated with untreated or unstable heart conditions affecting QT, or any QT-prolonging medicines.

• Kryxana 200mg Tablet tablets should be stored at room temperature between 20°C and 25°C (68°F and 77°F).
• It should be kept dry and protected from light and moisture.
• The tablets should be kept in the original packaging with the cap tightly closed.
• Keep it out of reach from children and pets.
• It is important to get the dose at the scheduled time. If you missed the schedule, contact your doctor and ask them about how to reschedule the dose.

The recommended dose is 600 mg daily, which equates to three 200 mg film-coated tablets, taken orally once a day for 21 consecutive days followed by a 7-day break, completing a 28-day cycle. Tablets should be swallowed whole with or without food, preferably at the same time each morning to maintain consistent levels. Dose may be reduced to 400 mg or 200 mg in case of intolerance or drug interactions.

Side Effects:

Side effects are unwanted symptoms caused by medicines. Even though all drugs cause side effects, not everyone gets them.

Serious:

• QT interval prolongation

• Hepatobiliary toxicity (liver enzyme elevations)

• Pneumonitis/interstitial lung disease

• Severe neutropenia (low white blood cell counts)

Common:

• Neutropenia

• Nausea, diarrhea, constipation

• Vomiting

• Fatigue, headache

• Hair thinning or loss

• Rash, cough

• Maintain routine blood tests (CBC, liver, ECG) throughout treatment cycles.

• Store the drug in its original packaging to ensure integrity and stability.

• Use daily pill pack reminders and avoid broken or damaged tablets.

• Inform your doctor promptly if you notice signs of infection, breathing difficulties, severe fatigue, or heart rhythm symptoms.

• Never share medication and always follow physician instructions for dose adjustments.
