Introduction
Semasize 0.25 mg is a solution for injection containing semaglutide 0.25 mg in 0.19 mL. It is supplied as a single-use prefilled syringe for administration under the skin.
Semaglutide belongs to a group of medicines called GLP-1 receptor agonists. Alkem's semaglutide injection range is approved in India for the management of type 2 diabetes mellitus and chronic weight management as an adjunct to diet and exercise.
Uses of Semasize 0.25 mg
Semasize 0.25 mg may be used for:
Management of type 2 diabetes mellitus along with diet and exercise
Chronic weight management as an adjunct to a reduced-calorie diet and increased physical activity
The doctor will decide the appropriate indication, dose and duration of treatment.
How Semasize 0.25 mg Works
Semasize 0.25 mg contains semaglutide, which mimics the action of GLP-1, a naturally occurring hormone involved in blood glucose and appetite regulation.
It increases insulin release when blood glucose is elevated and reduces glucagon release. It also slows gastric emptying and can reduce appetite. Together, these effects help improve blood glucose control and support weight management when combined with appropriate diet and physical activity.
Interactions of Semasize 0.25 mg with Other Medicines
Tell your doctor about all medicines you take before starting Semasize 0.25 mg.
Insulin or sulfonylureas: Using semaglutide with these medicines can increase the risk of low blood sugar. Your doctor may adjust their doses.
Oral medicines: Semaglutide delays gastric emptying and may affect the absorption of some oral medicines.
Warfarin or other coumarin medicines: Closer monitoring of blood clotting may be required when semaglutide treatment is started.
Other semaglutide-containing medicines or GLP-1 receptor agonists: Do not combine them with Semasize unless specifically advised by your doctor.
Storage Conditions
Store Semasize 0.25 mg in a refrigerator at 2°C to 8°C.
Do not freeze.
Keep it out of the reach of children.
Keep the medicine protected from light.
Follow the storage instructions on the product packaging and do not use the medicine if it has been frozen.
How to Use
Semasize 0.25 mg is given as a subcutaneous injection, meaning it is injected under the skin.
It is generally administered once a week, on the same day each week, as prescribed by your doctor.
It can be injected into the abdomen, thigh or upper arm.
The 0.25 mg dose is generally used during the initial phase of semaglutide treatment to help the body adjust to the medicine. Your doctor may increase the dose gradually depending on your response and tolerability.
The prefilled syringe is intended for single use. Do not reuse or share it.
Semasize can generally be administered with or without food.
Use only the dose prescribed by your doctor. Do not change the dose or frequency on your own.
Safety Advice
Pregnancy
Semaglutide should not be used during pregnancy. If you become pregnant while using Semasize, contact your doctor. Semaglutide should be stopped at least 2 months before a planned pregnancy because it has a prolonged elimination period.
Breast Feeding
Semaglutide is not recommended during breastfeeding because its safety in breastfed infants has not been established.
Lungs
Tell your doctor about any existing lung condition before starting Semasize. Seek medical attention if you develop difficulty breathing or swelling of the face or throat.
Liver
Inform your doctor if you have liver disease before using Semasize. Your doctor will assess whether the medicine is suitable for you.
Alcohol
Alcohol may worsen nausea, vomiting and other gastrointestinal symptoms. It can also affect blood glucose control, particularly when Semasize is used with other medicines that lower blood sugar.
Driving
Semaglutide itself is not expected to affect the ability to drive. However, dizziness or low blood sugar may affect driving ability, particularly when Semasize is used with insulin or a sulfonylurea.
Side Effects
Semasize 0.25 mg can cause gastrointestinal side effects, particularly during the early stages of treatment.

Serious side effects
Contact your doctor promptly if you develop severe or persistent symptoms such as:
Severe, persistent abdominal pain, which may indicate pancreatitis
Persistent vomiting or diarrhoea with signs of dehydration
Symptoms of low blood sugar, especially when used with insulin or sulfonylureas
Sudden changes in vision
Symptoms of a serious allergic reaction, such as swelling of the face or throat or difficulty breathing
Symptoms suggesting gallbladder problems, such as severe upper abdominal pain
Expert Tips
Use Semasize exactly as prescribed by your doctor.
The 0.25 mg dose is generally used at the beginning of treatment. Do not increase the dose yourself.
Take your injection on the same day each week to maintain a consistent routine.
Rotate injection sites between the abdomen, thigh and upper arm.
Do not inject Semasize into a vein or muscle.
The prefilled syringe is for single use only. Do not reuse or share it.
Drink enough fluids, particularly if you experience vomiting or diarrhoea.
Tell your doctor if you are taking insulin or a sulfonylurea because the risk of low blood sugar may increase.
Inform your doctor if you have a history of pancreatitis, gallbladder problems, severe stomach-emptying problems or diabetic eye disease.
If you are planning a pregnancy, discuss stopping Semasize with your doctor at least 2 months beforehand.
