Sembolic may be used for:

• Improving blood glucose control in adults with insufficiently controlled type 2 diabetes mellitus

• Chronic weight management in adults, along with a reduced-calorie diet and increased physical activity

• Supporting management of type 2 diabetes along with appropriate diet and exercise

The doctor decides the appropriate indication, dose and duration of treatment.

Sembolic contains semaglutide, which mimics the action of GLP-1, a hormone involved in blood glucose and appetite regulation.

It increases insulin release when blood glucose levels are elevated and reduces glucagon secretion. It also slows gastric emptying and can reduce appetite. These effects help improve blood glucose control and support weight management when used with appropriate diet and physical activity.

Tell your doctor about all medicines you take before starting Sembolic.

• Insulin or sulfonylureas: Using semaglutide with these medicines can increase the risk of low blood sugar. Their doses may need adjustment.

• Oral medicines: Semaglutide can delay gastric emptying and may affect the absorption of some medicines taken by mouth.

• Warfarin or other coumarin medicines: Your doctor may recommend closer monitoring of blood clotting when semaglutide treatment is started.

• Other semaglutide-containing medicines or GLP-1 receptor agonists: Do not use them together with Sembolic unless specifically advised by your doctor.

• Store Sembolic according to the storage instructions provided with the product packaging.

• Do not freeze the cartridge.

• Protect the medicine from excessive heat and light.

• Keep it out of reach of children.

• Follow the product-specific instructions for storage of the cartridge after it has been inserted into the reusable pen.

• Sembolic is administered as a subcutaneous injection using the compatible reusable pen device.

• Semaglutide is generally administered once weekly.

• It may be injected into the abdomen, thigh or upper arm.

• Do not inject into a vein or muscle.

• The 15 mg/3 mL cartridge contains enough semaglutide to provide different prescribed dose strengths during dose escalation and maintenance treatment. The clinical formulation was designed to deliver doses of 0.25 mg, 0.5 mg, 1 mg, 1.7 mg and 2.4 mg through the same pen platform.

• Use only the dose selected and prescribed by your doctor.

• Do not change the dose or injection frequency yourself.

• Follow the instructions supplied with the reusable pen for inserting the cartridge, selecting the dose and administering the injection.

Sembolic may cause gastrointestinal side effects, particularly when treatment is started or the dose is increased.

Common side effects

• Nausea

• Decreased appetite

• Indigestion

• Headache

• Dizziness

Serious side effects

Contact your doctor promptly if you develop:

• Severe or persistent abdominal pain, which may indicate pancreatitis

• Persistent vomiting or diarrhoea with signs of dehydration

• Symptoms of low blood sugar, particularly when using insulin or a sulfonylurea

• Sudden changes in vision

• Symptoms of a serious allergic reaction, such as swelling of the face or throat or difficulty breathing

• Severe abdominal pain that may indicate gallbladder problems

• Use Sembolic exactly as prescribed by your doctor.

• Take the injection on the same day each week.

• Use the compatible reusable pen according to the manufacturer's instructions.

• Do not change the prescribed dose yourself.

• Rotate injection sites between the abdomen, thigh and upper arm.

• Do not inject into a vein or muscle.

• Drink enough fluids, particularly if you experience vomiting or diarrhoea.

• Tell your doctor if you use insulin or a sulfonylurea because your risk of low blood sugar may increase.

• Inform your doctor if you have a history of pancreatitis, gallbladder disease, diabetic eye disease or severe gastrointestinal problems.

• Tell your healthcare team that you use semaglutide before undergoing general anaesthesia or deep sedation.
